目的探讨亚甲基四氢叶酸还原酶(MTHFR)基因多态性与1,3-丁二烯作业工人DNA损伤易感性的关系。方法选取255名1,3-丁二烯作业工人和60名行政人员作为研究对象,采用碱性彗星试验评价个体外周血淋巴细胞DNA损伤,聚合酶链反应-限制性片段长度多态性方法分析MTHFR基因多态性。结果暴露组中MTHFR1298位点AA基因型个体彗星尾距(4.7)显著高于AC(4.5)或AC+CC(4.4)基因型个体。而MTHFR基因单体型分析则未发现有显著性差异。结论 1,3-丁二烯作业工人MTHFR A1298C位点的多态性可能影响其染色体损伤的易感性。
Objective To investigate the relationship between methylenetetrahydrofolate reductase (MTHFR) gene polymorphism and DNA damage susceptibility of workers in 1,3-butadiene. Methods A total of 255 1,3-butadiene laborers and 60 administrative staff were enrolled in this study. Alkaline comet assay was used to evaluate the DNA damage of peripheral blood lymphocytes and the polymerase chain reaction-restriction fragment length polymorphism MTHFR gene polymorphism. Results The comet tail distance (4.7) in MTHFR1298 locus AA genotype was significantly higher than that in AC (4.5) or AC + CC (4.4) genotype individuals in the exposed group. MTHFR gene haplotype analysis found no significant difference. Conclusion The polymorphism of MTHFR A1298C in 1,3-butadiene workers may affect the susceptibility of chromosome damage.
