目的:研究胆红素的吸附行为,探索最佳分析条件。方法:利用石英晶体微天平研究磷酸盐缓冲溶液(PBS)中胆红素在卵磷脂双层膜的吸附行为,石英晶体震荡频率下降越多,表示吸附量越多。考察了溶液pH值、离子强度和温度的影响,并与巯基苯胺膜和巯基苯胺/卵磷脂修饰膜进行了比较。结果:溶液pH为8.0时,频率变化-780Hz;溶液中离子强度增加一倍,频率变化-76Hz;10℃、28℃时,频率变化分别为-709Hz和-374Hz。结论:卵磷脂双层膜在碱性条件下有最大吸附;离子强度也会增加胆红素在膜上的吸附,但是影响没有其它因素大;温度会显著的影响胆红素的吸附,低温时吸附量明显增加;不同膜上胆红素的吸附量不同,其中硫醇/卵磷脂修饰膜上最大。
Objective: To study the adsorption behavior of bilirubin and explore the best analytical conditions. Methods: The adsorption behavior of bilirubin in lecithin bilayer membrane in phosphate buffered saline (PBS) was studied by using quartz crystal microbalance. The more the quartz crystal oscillation frequency decreased, the more the adsorption capacity was. The effects of pH value, ionic strength and temperature of the solution were investigated. The results were compared with the modified films of mercaptoaniline and mercaptoaniline / lecithin. Results: When the pH of the solution was 8.0, the frequency changed -780Hz. The ionic strength of the solution doubled and the frequency changed -76Hz. At 10 ℃ and 28 ℃, the frequency change was -709Hz and -374Hz respectively. CONCLUSION: The lecithin bilayer membrane has the maximum adsorption under alkaline conditions; the ionic strength also increases the adsorption of bilirubin on the membrane, but no other factors are affected; the temperature will significantly affect the adsorption of bilirubin, and at low temperature The amount of adsorption increased significantly; the adsorption amount of bilirubin on different membranes was different, of which the thiol / lecithin membrane was the largest.
