在高血压患者中研究血管紧张素原 (AGT)基因M2 35T多态性、等位基因分离及与细胞因子的关系。采用聚合酶链反应结合内切酶消化进行AGT基因型分析。用双抗体夹心ELISA酶联法测定细胞因子 (IL 1、IL 6、TNF)。分析高血压病组和对照组的血管紧张素原 (AGT)基因 2 35位氨基酸残基的基因型及等位基因频率 ,并分析细胞因子对AGT基因M2 35T变异的影响。结果表明 :高血压组中AGT 2 35为TT型 38例 (5 5 88% ) ,TM型 2 4例(35 2 9% ) ,MM型 6例 (8 82 % ) ,其T型和M型等位基因频率分别为 73 5 3%和 2 6 4 7%。对照组中TT型 2 8例(47 4 6 % ) ,TM型 2 5例 (42 37% ) ,MM型 6例 (10 17% ) ,T型和M型等位基因频率分别为 6 8 6 5 %和 31 35 %。在AGT 2 35T等位基因型组中 ,高血压病组IL 1、IL 6、TNF浓度显著高于对照组。在AGT 2 35M等位基因型组中 ,TNF浓度高于对照组。无论大于 6 0岁或小于 6 0岁的高血压病与相应年龄对照组比较 ,IL 1、IL 6、TNF浓度均显著升高。而在高血压病或对照组内 ,大于 6 0岁组与小于 6 0岁组之间比较 ,IL 1、IL 6、TNF浓度均无显著性差异。说明IL 1、IL 6、TNF升高与疾病有关而与年龄无关。研究结论 :高血压病AGT 2 35TT型和T等位基因频率高于健康对照。AGT 2 35TT型和T等位基因频率?
To study the relationship between M235T polymorphism of angiotensinogen (AGT) gene, allelic segregation and cytokines in hypertensive patients. AGT genotyping was performed by polymerase chain reaction and endonuclease digestion. Cytokines (IL 1, IL 6, TNF) were determined by double antibody sandwich ELISA. The genotypes and allele frequencies of amino acid residues of 235 of the angiotensinogen (AGT) gene in hypertension group and control group were analyzed. The effects of cytokines on the M235T mutation of AGT gene were analyzed. The results showed that there were 38 TT patients (55.8%), 24 (35.29%) TM patients and 6 (8.82%) MM patients in hypertensive group. The T-type and M-type Allele frequencies were 73.5% and 26.47%, respectively. The control group had 28 TT genotypes (47 4 6%), 25 TM genotypes (42 37%) and 6 MM genotypes (10 17%). The frequencies of T genotypes and M genotypes were 6 8 6 5% and 31 35%. In AGT 2 35T allele group, the concentrations of IL 1, IL 6 and TNF in hypertension group were significantly higher than those in control group. In the AGT 2 35M allele group, the TNF concentration was higher than that of the control group. The levels of IL-1, IL-6 and TNF in hypertensive patients older than 60 years or younger were significantly higher than those in the corresponding age-matched controls. However, no significant differences were found in the levels of IL-1, IL-6 and TNF between the hypertensive group and the control group over 60 years of age and less than 60 years of age. That IL 1, IL 6, TNF and disease-related and has nothing to do with age. Conclusions: The frequency of AGT 2 35TT and T alleles in hypertensive patients is higher than that in healthy controls.
