目的研究疏肝清热健脾法对广泛性焦虑(GAD)大鼠脑内Papez环路区域的B细胞白血病/淋巴瘤-2(bcl-2)、B细胞白血病/淋巴瘤-2相关蛋白(bax)2种蛋白表达的影响。方法将40只雄性Wistar大鼠随机分为正常组、模型组、中药治疗组和西药对照组4组,每组10只大鼠。模型组、治疗组、对照组采用不确定空瓶饮水刺激方法制备广泛性焦虑大鼠模型。治疗组和对照组分别以丹栀逍遥散(10.5 g/kg)和盐酸丁螺环酮(2 mg/kg)水溶液灌胃7 d,每天1次。模型组蒸馏水灌胃7 d。取脑组织,western blot法检测大鼠脑内海马和乳头体组织bax和bcl-2的含量。结果与正常组比较,模型组海马和乳头体组织的bax含量显著增加,差异有统计学意义(P<0.05或P<0.01),bcl-2的含量有较明显的下降,差异有统计学意义(P<0.01);与模型组相比,治疗组海马及乳头体组织的bax表达均有降低,差异有统计学意义(P<0.05或P<0.01),bcl-2的表达均有增加,差异有统计学意义(P<0.01)。结论以丹栀逍遥散为代表的疏肝清热健脾法能够降低GAD大鼠脑内海马和乳头体组织bax的水平,升高bcl-2的水平,降低bax/bcl-2值,干预和抑制Papez环路区域的细胞凋亡,从而改善焦虑症状。
Objective To study the effects of “Shugan Qingre Jianpi Method” on B-cell leukemia / lymphoma-2 (Bcl-2), B cell leukemia / lymphoma-2 associated protein (Bax) in the Papez loop region of the brain of generalized anxiety (GAD) ) Two kinds of protein expression. Methods Forty male Wistar rats were randomly divided into normal group, model group, TCM treatment group and western medicine control group, 4 rats in each group. Results Compared with the normal group, the content of bax in hippocampus and papillary tissues in model group increased significantly (P <0.05 or P <0.01), and the content of bcl-2 decreased significantly with statistical significance (P <0.01). Compared with the model group, the expression of bax in the hippocampus and the papillary tissue in the treatment group decreased significantly (P <0.05 or P <0.01), the expression of bcl-2 increased, The difference was statistically significant (P <0.01). Conclusion Shugan Qingre Decoction, represented by Danzhi Xiaoyao Powder, can decrease the level of bax, increase the level of bcl-2 and decrease the bax / bcl-2 levels in the hippocampus and papillary tissues of GAD rats, intervene and inhibit Papez loop area of the cell apoptosis, thereby improving anxiety symptoms.
