由于常规福尔马林固定和石蜡包理过程造成的抗原封闭与结构改变,限制了一些抗体在免疫组织化学中的应用。为了恢复组织的抗原性,提高免疫组织化学的检测阳性率,我们应用高压锅,微波炉和胰蛋白酶分别对石蜡切片进行处理。结果显示,一些抗体过去只能用于冰冻切片,经前处理后可用于石蜡切片,一些抗体的敏感性增强,经高压锅处理的免疫染色重复性较微波炉处理得好,个别抗体适用于酶消化。总之,在免疫组织化学染色过程中应根据抗原抗体的特性选用适当的前处理方法。
Due to the blocking of the antigen and the structural changes caused by the routine formalin fixing and the paraffin waxing process, the application of some antibodies in immunohistochemistry is limited. In order to restore the antigenicity of the tissue and increase the positive rate of immunohistochemistry, paraffin sections were treated with autoclave, microwave and trypsin respectively. The results showed that some antibodies were only used in frozen sections and could be used for paraffin sections after pretreatment. Some antibodies were more sensitive. Immunoblotting by autoclave was more reproducible than microwave oven. Individual antibodies were suitable for enzymatic digestion. In short, in immunohistochemical staining should be based on the characteristics of antigen-antibody selection appropriate pre-treatment method.
