目的探讨急性心肌缺血大鼠心肌组织中肾素-血管紧张素系统(RAS)的表达变化。方法通过结扎大鼠冠状动脉左前降支复制急性心肌缺血模型。采用RT-PCR方法测定心肌组织中血管紧张素原(AGT)、血管紧张素转换酶(ACE)、血管紧张素1型受体(AT1)/血管紧张素2型受体(AT2)及水通道蛋白4(AQP4)mRNA的表达。结果与假手术组相比,急性心肌缺血组大鼠心肌组织中AGT mRNA的变化不明显,ACE及AT1 mRNA的表达明显降低(P<0.05),AT2及AQP4 mRNA的表达明显升高(P<0.05)。结论局部RAS及AQP4参与了急性心肌缺血的过程。
Objective To investigate the expression of renin-angiotensin system (RAS) in myocardium of acute myocardial ischemia rats. Methods Acute myocardial ischemia model was induced by ligation of left anterior descending coronary artery in rats. The levels of angiotensinogen (AGT), angiotensin converting enzyme (ACE), angiotensin type 1 receptor (AT1) / angiotensin type 2 receptor (AT2) and water channels in myocardium were measured by RT- Protein 4 (AQP4) mRNA expression. Results Compared with the sham operation group, the changes of AGT mRNA in acute myocardial ischemia group were not obvious, the expressions of ACE and AT1 mRNA were significantly decreased (P <0.05), and the expressions of AT2 and AQP4 mRNA were significantly increased (P <0.05). Conclusion Local RAS and AQP4 are involved in the process of acute myocardial ischemia.
