目的:探讨表皮生长因子样结构域7(EGFL7)在恶性脑膜瘤细胞及组织的表达及意义。方法:体外培养人恶性脑膜瘤细胞株(IOMM-LEE、CH157-MN)及正常人脑膜细胞系(HMC-1400)。采用实时荧光定量PCR(qRT-PCR)、反转录PCR(RT-PCR)、蛋白免疫印迹(Western blot)等方法检测EGFL7 mRNA及EGFL7蛋白在细胞株IOMM-LEE、CH157-MN和细胞系HMC-1400中表达,检测25例恶性脑膜瘤肿瘤组织及相应的瘤周正常蛛网膜组织、侵袭的颅骨组织中EGFL7 mRNA及EGFL7蛋白的表达水平。结果:qRT-PCR、RT-PCR及Western blot结果显示人恶性脑膜瘤细胞株IOMM-LEE和CH157-MN中的EGFL7 mRNA及蛋白表达水平均高于正常人脑膜细胞系HMC-1400(n P<0.05)。恶性脑膜瘤组织中的EGFL7 mRNA及蛋白表达水平均高于相应瘤周正常蛛网膜组织(n P<0.05);EGFL7的蛋白和mRNA表达水平在侵袭颅骨组织、原发恶性脑膜瘤组织、瘤周正常蛛网膜组织中的表达水平依次递减(n P<0.05)。n 结论:EGFL7在恶性脑膜瘤组织及细胞系中呈高表达,且其高表达可能与恶性脑膜瘤侵袭相关。“,”Objective:To investigate the expression and significance of epidermal growth factor-like domain 7 (EGFL7) in malignant meningioma.Methods:Human malignant meningeal cell lines (IOMM-LEE, CH157-MN) and normal meningeal cells (HMC-1400) were cultured in n vitro. The expressions of EGFL7 mRNA and protein in cells were detected by real time fluorescent quantitative polymerase chain reaction (qRT-PCR), reverse polymerase chain reaction (RT-PCR), and Western blot. The expression levels of EGFL7 mRNA and protein in 25 cases of malignant meningioma, the corresponding normal arachnoid tissue around the tumor and the invasive skull tissue were detected.n Results:The results of qRT-PCR, RT-PCR and Western blot showed that the expression levels of EGFL7 mRNA and protein in human malignant meningeal cell lines (IOMM-LEE, CH157-MN) were higher than those in normal meningeal cells (HMC-1400) (n P<0.05). The expression levels of EGFL7 mRNA and protein in malignant meningiomas were higher than those in normal arachnoid tissues around the tumors (n P<0.05); the expression levels of EGFL7 mRNA and protein in invasive cranial tissues, primary malignant meningiomas and normal arachnoid tissues around the tumors decreased in turn (n P<0.05).n Conclusions:EGFL7 is highly expressed in malignant meningioma tissues and cell lines, and its high expression may be associated with malignant meningioma invasion.
