目的:观察血小板衍化生长因子(PDGF)、氧化低密度脂蛋白内皮受体-1(LOX-1)、转移生长因子(TGF-β)等因子在糖尿病肾病的发生、发展中的作用。方法:用反转录半定量聚合酶链反应(RT-PCR)法测定18只STZ模型鼠(DN组)及6只对照鼠(NC组)的PDGF、LOX-1、TGF-β等基因表达以及mRNA含量的变化。结果:DN组大鼠肾皮质PDGFmRNA、LOX-1mRNA、TGF-βmRNA基因表达较NC组明显增高;(P<0.01)。结论:在糖尿病后期,PDGF、LOX-1、TGF-β生成增加,参与并加剧了糖尿病肾脏的病变。
Objective: To observe the effect of PDGF, LOX-1 and TGF-β on the occurrence and development of diabetic nephropathy. Methods: The mRNA expressions of PDGF, LOX-1 and TGF-β in 18 STZ model rats (DN group) and 6 control rats (NC group) were determined by reverse transcription semi-quantitative polymerase chain reaction As well as the change of mRNA content. Results: The expressions of PDGFmRNA, LOX-1mRNA and TGF-βmRNA in renal cortex of DN group were significantly higher than those in NC group (P <0.01). Conclusion: In the later stage of diabetes, PDGF, LOX-1 and TGF-β are increased, and they are involved in and aggravate the pathological changes of diabetic nephropathy.
