目的探讨胭脂树橙在诱导白血病K562细胞凋亡过程中对PPARγ蛋白表达的影响。方法用胭脂树橙作用于K562细胞一定时间后,通过台盼蓝拒染法观察K562细胞生长曲线,MTT法检测其对细胞的抑制率,Hoechst 33258染色后,荧光显微镜下观察细胞核形态;流式细胞仪检测细胞的凋亡率;采用Western-blotting技术从蛋白分子水平上检测胭脂树橙对PPARγ蛋白表达的影响。结果胭脂树橙显著抑制K562细胞的增殖并呈浓度依赖关系。电泳的结果显示一定浓度的胭脂树橙能够上调PPARγ蛋白的表达,进而可能诱导K562细胞的凋亡。结论PPARγ蛋白表达的上调可能是胭脂树橙抑制K562细胞增殖诱导凋亡的原因之一,其中,胭脂树橙具有PPARγ非特异配体的作用。
Objective To investigate the effect of bixin on the expression of PPARγ protein in the process of leukemia K562 cell apoptosis. Methods The K562 cells were treated with bixin orange for a certain period of time. The growth curve of K562 cells was observed by trypan blue exclusion method. The inhibition rate of K562 cells was determined by MTT assay. The morphology of K562 cells was observed under Hoechst 33258 staining. The apoptosis rate of cells was detected by cytometry. Western blotting was used to detect the effect of bixin on the expression of PPARγ protein at the protein level. Results Catharanthi orange significantly inhibited the proliferation of K562 cells in a concentration-dependent manner. The results of electrophoresis showed that a certain concentration of bixin could up-regulate the expression of PPARγ protein, which may induce the apoptosis of K562 cells. Conclusions The up-regulation of PPARγ protein expression may be one of the reasons that the proliferation of Kakihimes could inhibit the proliferation of K562 cells, in which birch orange has the function of PPARγ nonspecific ligand.
