目的建立CB17 SCID小鼠KATOⅢ胃癌皮下肿瘤模型,为胃癌的临床前研究提供更有效的动物模型。方法将KATOⅢ细胞接种至雌性CB17 SCID小鼠皮下,待肿瘤长至约200mm3时,取出肿瘤切成约30mm3小块后再次接种到雌性CB17 SCID小鼠皮下,观察肿瘤的生长情况,并研究肿瘤的组织病理学特征。结果该肿瘤模型成瘤率为100%,肿瘤生长情况良好,组织病理学检查完全符合胃癌细胞的形态学特征。结论通过肿瘤组织块异体移植的方法建立KATOⅢ胃癌皮下肿瘤模型可以缩短肿瘤的生长时间,为临床前胃癌的研究提供可靠的动物模型。
Objective To establish a KATO Ⅲ gastric cancer subcutaneous tumor model in CB17 SCID mice and to provide a more effective animal model for the preclinical study of gastric cancer. Methods KATO Ⅲ cells were inoculated subcutaneously into female CB17 SCID mice. When the tumors grew to about 200 mm 3, the tumors were removed and cut into small pieces of about 30 mm 3. The cells were inoculated subcutaneously into female CB17 SCID mice to observe the growth of the tumors and to study the effects of Histopathological features. Results The tumor model had a tumorigenic rate of 100% and good tumor growth. Histopathological examination was completely consistent with the morphological characteristics of gastric cancer cells. Conclusion The establishment of KATO Ⅲ subcutaneous tumor model by tumor allograft can shorten tumor growth time and provide a reliable animal model for the study of precancerous gastric cancer.
