核磁共振代谢组学法研究大蒜辣素对大鼠的作用机制。以生物核磁共振技术结合偏最小二乘-辨别分析法(PLS-DA)分析大鼠腹腔注射大蒜辣素后尿液内源性代谢产物的变化,研究大蒜辣素对正常大鼠代谢过程的影响。给药后,尿样中三羧酸循环中间产物柠檬酸、α-酮戊二酸、琥珀酸含量呈现出先升高后逐渐恢复的趋势,酮体水平也有所上升;而葡萄糖、乳酸、丙氨酸、马尿酸、氮氧三甲胺和牛磺酸水平略有降低。偏最小二乘分析表明,给药组与对照组的代谢谱有明显差异。大蒜辣素给药后对正常大鼠代谢过程具有显著影响,代谢组学研究有助于认识大蒜的作用机制。
Study on Mechanism of Alloxan in Rats by Nuclear Magnetic Resonance Metabolomics. The changes of urinary endogenous metabolites in rats after intraperitoneal injection of allicin were analyzed by bio-nuclear magnetic resonance and PLS-DA, and the effects of allicin on the metabolism of normal rats . After administration, citric acid, α-ketoglutaric acid and succinic acid in urine samples showed a trend of first rising and then recovering, and the level of ketone body also increased. However, glucose, lactate and ammonia Acid, hippuric acid, triamcinolone and taurine slightly decreased. Partial least squares analysis showed that there was a significant difference in the metabolic profiles between the treated group and the control group. Allicin administration has a significant effect on the metabolism of normal rats, metabolomics research helps to understand the mechanism of garlic.
