目的 研究小鼠骨髓基质细胞K+通道的特性。方法 采用膜片钳细胞附式和内面向外式的单通道记录方式记录原代培养的小鼠骨髓基质K+通道电流。结果 骨髓基质细胞膜上存在一类单通道电导为(16.7±1.4)pS的K+通道,通道电流活动在75 s内不失活。随着去极化程度的增大,通道的开放时间τo2延长,关闭时间τc2缩短,通道开放概率增加。结论 骨髓基质细胞膜上存在一性质类似于延迟整流的K+通道,提示后者在骨髓基质细胞信号转导中起作用。
Objective To study the characteristics of K + channels in mouse bone marrow stromal cells. Methods Primary cultured mouse bone marrow stromal K + channel currents were recorded using patch-clamp cell attachment and inside-out single-channel recording. Results There was a K + channel with single-channel conductance (16.7 ± 1.4) pS on the membrane of bone marrow stroma, and the channel current activity was not inactivated within 75 s. As the degree of depolarization increases, the channel opening time τo2 extends, the closing time τc2 shortens, and the channel opening probability increases. Conclusion There is a K + channel in bone marrow stromal cell membrane with delayed rectification, suggesting that the latter plays a role in signal transduction of bone marrow stromal cells.
