目的:从定量角度探讨p53、ras p21和PCNA在非小细胞肺癌中异常表达的特点及与肿瘤分型、分级和局部淋巴结癌转移的关系及其生物学意义。方法:应用抗p53、ras p21、PCNA单克隆抗体对65例非小细胞肺癌进行LSAB免疫组化法染色。结果:ras p21表达腺癌高于鳞癌(P<0.05),在全部病例中ras p21阳性表达Ⅰ级分别高于Ⅱ级和Ⅲ级(P<0.05),腺癌Ⅰ级ras p21表达高于Ⅲ级(P<0.05)。PCNA表达鳞癌显著高于腺癌(P<0.01),在全部病例中其表达Ⅲ级高于Ⅰ级,在鳞癌中PCNA表达Ⅲ级分别高于Ⅰ级和Ⅱ级(P<0.05)。而p53表达与肺癌分型、分级无关。等级相关分析表明p53与PCNA在肺癌中的表达呈显著正相关(总病例rs=0.2591,P<0.05,腺癌rs=0.4275,P<0.05。结论:(1)ras p21、PCNA表达与肺癌分型、分化有关,检测非小细胞肺癌 ras p21、PCNA的表达有助于肺癌分型、分化的判断。(2)肺癌中p53蛋白与PCNA这两种分子可能具有协同调节细胞增殖和分化的作用。
OBJECTIVE: To investigate the characteristics of abnormal expression of p53, ras p21 and PCNA in non-small cell lung cancer and their relationship with tumor type, grade, metastasis of regional lymph nodes and their biological significance. METHODS: LSAB immunohistochemical staining was performed on 65 cases of non-small cell lung cancer with anti-p53, ras p21, and PCNA monoclonal antibodies. Results: The expression of ras p21 in adenocarcinoma was higher than that in squamous cell carcinoma (P<0.05). In all cases, the positive expression of ras p21 was higher than that of grade II and III (P<0.05). The expression of ras p21 in grade I of adenocarcinoma was higher than that of grade II and III. Grade III (P < 0.05). PCNA-expressed squamous cell carcinoma was significantly higher than that of adenocarcinoma (P<0.01). In all cases, its expression level III was higher than that of grade I. PCNA expression level III in squamous cell carcinoma was higher than that of grade I and II (P<0.05). However, p53 expression has nothing to do with the classification or grade of lung cancer. The hierarchical correlation analysis showed that there was a significant positive correlation between p53 and PCNA expression in lung cancer (rs=0.2591 in total cases, P<0.05, rs=0.4275 in adenocarcinomas, P<0.05. Conclusion: (1) Expression of ras p21, PCNA and lung cancer The expression of ras p21 and PCNA in non-small cell lung cancer can contribute to the classification and differentiation of lung cancer.(2) The two molecules p53 protein and PCNA may have a synergistic effect on the regulation of cell proliferation and differentiation in lung cancer. .
