目的探讨Wnt2和β-catenin在乳腺癌及其配对癌旁乳腺组织中的表达与临床意义。方法采用RT-PCR法检测7份乳腺癌及其配对癌旁乳腺组织中Wnt2和β-catenin基因mRNA的转录水平;免疫组织化学方法检测40份浸润性乳腺导管癌及其配对癌旁乳腺组织中Wnt2和β-catenin蛋白的表达水平;分析Wnt2和β-catenin与乳腺癌临床病理特征的相关性及二者联合检测与乳腺癌恶性程度及淋巴结转移的相关性。结果乳腺癌组织中Wnt2和β-catenin基因mRNA的相对转录水平均显著高于配对癌旁乳腺组织(P<0.05)。Wnt2在配对癌旁乳腺组织细胞质中呈弱表达,在浸润性乳腺导管癌组织细胞质中呈高表达,乳腺癌Wnt2阳性表达率(77.5%)显著高于配对癌旁乳腺组织(15.0%)(P<0.05);配对癌旁乳腺组织中β-catenin均表达于细胞膜中,在浸润性乳腺导管癌组织细胞质及细胞核中均有表达,异常表达率为67.5%(27/40)。Wnt2和β-catenin的表达与浸润性乳腺导管癌患者年龄无关(P>0.05),与乳腺癌临床分期、病理组织学分级及淋巴结转移有关(P<0.05)。Wnt2和β-catenin均为阳性表达者的肿瘤恶性程度和淋巴结转移的发生率显著高于两者均为阴性或两者之一为阳性表达者(P<0.05)。结论 Wnt2和β-catenin表达在乳腺癌发生和侵袭转移中可能起重要作用,本实验为进一步探讨Wnt2和β-catenin在乳腺癌发病机制中的作用奠定了基础。
Objective To investigate the expression and clinical significance of Wnt2 and β-catenin in breast cancer and its adjacent tissues. Methods RT-PCR was used to detect the transcriptional levels of Wnt2 and β-catenin mRNA in 7 breast cancer tissues and paired paired adjacent breast tissues. Immunohistochemistry was used to detect the expression of mRNA of Wnt2 and β-catenin in 40 breast cancer tissues and adjacent tissues Wnt2 and β-catenin protein levels in breast cancer; analyze the correlation between Wnt2 and β-catenin and the clinicopathological features of breast cancer; and the correlation between Wnt2 and β-catenin and the malignant degree and lymph node metastasis of breast cancer. Results The relative transcriptional levels of Wnt2 and β-catenin mRNA in breast cancer tissues were significantly higher than those in paired para-cancerous tissues (P <0.05). Wnt2 was weakly expressed in the cytoplasm of matched paracancer breast tissues and highly expressed in the cytoplasm of invasive ductal breast carcinomas. The positive rate of Wnt2 in breast cancer tissues (77.5%) was significantly higher than that in matched paracancer breast tissues (15.0%) (P <0.05). The expression of β-catenin in the paired adjacent breast cancer tissues was both in the cell membrane and in the cytoplasm and nucleus of invasive ductal breast cancer tissues. The abnormal expression rate was 67.5% (27/40). The expression of Wnt2 and β-catenin was not related to the age of infiltrating breast ductal carcinoma (P> 0.05), but correlated with the clinical staging, pathological grade and lymph node metastasis (P <0.05). The malignant degree and the incidence of lymph node metastasis in both Wnt2 and β-catenin were significantly higher than those in both of the two groups (P <0.05). Conclusion The expression of Wnt2 and β-catenin may play an important role in the genesis and invasion and metastasis of breast cancer. This study lays the foundation for further exploring the role of Wnt2 and β-catenin in the pathogenesis of breast cancer.
