目的探讨叉头转录因子(FOXO3a)、β-连环素(β-Catenin)、增殖细胞核抗原(PCNA)在乳腺癌发生、发展中的作用。方法选择我院手术切除的乳腺癌组织标本68份(肿瘤组)和乳腺纤维腺瘤组织标本20份(腺瘤组),采用免疫组化Envision两步法检测两组FOXO3a、β-Catenin、PCNA蛋白表达;采用Spearman等级相关分析法分析三指标间及与乳腺癌临床病理参数的相关性。结果肿瘤组、腺瘤组FOXO3a阳性表达率分别为60%9、0%,β-Catenin异常表达率分别为65%、25%,PCNA阳性表达率分别为69%3、5%,两两比较P均<0.05;FOXO3a阳性表达与乳腺癌临床分期、雌激素受体、腋窝淋巴结转移、预后相关,β-Catenin异常表达与乳腺癌组织学分级、腋窝淋巴结转移、临床分期相关,PCNA阳性表达与乳腺癌腋窝淋巴结转移、临床分期相关;乳腺癌组织中FOXO3a阳性表达与β-Catenin异常表达、PCNA阳性表达均呈负相关,β-Catenin异常表达与PCNA阳性表达呈正相关。结论 FOXO3a低表达、β-Catenin异常表达、PCNA高表达与乳腺癌的发生、发展相关。
Objective To investigate the roles of FOXO3a, β-Catenin and PCNA in the development and progression of breast cancer. Methods 68 cases of breast cancer (tumor group) and 20 cases of breast fibroadenoma (adenoma group) resected in our hospital were selected. The expression of FOXO3a, β-Catenin and PCNA Protein expression. Spearman rank correlation analysis was used to analyze the correlation between three indicators and clinicopathological parameters of breast cancer. Results The positive expression rates of FOXO3a in tumor group and adenoma group were 60%, 9%, respectively. The abnormal expression rates of β-Catenin were 65%, 25%, respectively. The positive rates of PCNA were 69%, 35% P <0.05; The positive expression of FOXO3a was correlated with the clinical stage, estrogen receptor, axillary lymph node metastasis and prognosis of breast cancer. The abnormal expression of β-Catenin correlated with histological grade, axillary lymph node metastasis, clinical stage, Breast cancer axillary lymph node metastasis, clinical stage related; breast cancer FOXO3a positive expression and β-Catenin abnormal expression, PCNA positive expression were negatively correlated, β-Catenin abnormal expression and PCNA positive expression was positively correlated. Conclusion FOXO3a low expression, abnormal expression of β-Catenin, high expression of PCNA associated with the occurrence and development of breast cancer.
