目的:观察黄芪多糖对孕早期蜕膜TLR9表达与功能的影响。方法:对孕早期蜕膜进行原代培养,将其分为两组,PBS对照组、CPG-ODN刺激组,用0、25、50、125、250mg/L黄芪多糖预处理两组蜕膜细胞48h,再分别用PBS、CPG-ODN处理24h,采用酶联免疫吸附试验方法检测上清液TNF-α、IL-6的分泌,用流式细胞仪检测蜕膜细胞TLR9的表达。结果:用CPG-ODN刺激蜕膜细胞后,TLR9荧光表达(1.85±0.014)、IL-6的分泌(26.80±13.91)pg/mL显著高于刺激前(0.680±0.276)、(10.34±2.36)pg/mL(P<0.05),TNF-α的分泌(11.57±1.040)pg/mL与刺激前(9.548±0.458)pg/mL无显著性差异(P>0.05);用25、50、125、250mg/L黄芪多糖预处理蜕膜细胞48h后,用CPG-ODN刺激细胞,TLR9的表达与TNF-α、IL-6的分泌与未用黄芪多糖预处理相比无显著性差异(P>0.05)。结论:Toll样受体9功能性表达于孕早期蜕膜,但黄芪多糖对TLR9及其介导的TNF-α、IL-6的分泌无显著性影响。
Objective: To observe the effect of astragalus polysaccharide on the expression and function of decidual TLR9 in early pregnancy. Methods: The decidua of the first trimester were primary cultured and divided into two groups: PBS control group and CPG-ODN stimulation group. The decidual cells were pretreated with 0, 25, 50, 125 and 250 mg / L APS 48h. The cells were treated with PBS and CPG-ODN for 24 h respectively. The secretion of TNF-α and IL-6 in the supernatant was detected by enzyme linked immunosorbent assay. The expression of TLR9 in decidual cells was detected by flow cytometry. Results: The expression of TLR9 (1.85 ± 0.014), IL-6 secretion (26.80 ± 13.91) pg / mL was significantly higher than that before stimulation (0.680 ± 0.276) and (10.34 ± 2.36) after stimulation with CPG- (P <0.05). The secretion of TNF-α (11.57 ± 1.040) pg / mL was not significantly different from that before stimulation (9.548 ± 0.458) pg / mL After being pretreated with 250mg / L APS for 48h, the expression of TLR9 and the secretion of TNF-α and IL-6 in CPG-ODN group were not significantly different from those of APS group (P> 0.05 ). CONCLUSION: Toll-like receptor 9 is functionally expressed in the decidua of early pregnancy, but APS has no significant effect on the secretion of TLR9, TNF-α and IL-6.
