目的:探讨不同浓度吡咯烷二硫氨基甲酸(PDTC)对高糖下大鼠成纤维细胞(NRK)中单核细胞趋化蛋白-1(MCP-1)表达的影响。方法:体外培养大鼠NRK细胞;葡萄糖孵育下分6组,正常对照组、高糖1组、高糖2组含葡萄糖分别为5.6、15和30mmol/L,干预组在葡萄糖30mmo/L基础上分别加入PDTC5、10和20μmol/L;RT-PCR检测24、48h后各组MCP-1mRNA表达水平,免疫细胞化学(ICC)法检测24、48h各组的MCP-1蛋白表达情况。结果:与正常对照组相比,高糖组MCP-1mRNA和蛋白表达水平显著升高(P<0.05),且呈剂量时间依赖性;而不同浓度PDTC干预后,MCP-1mRNA和蛋白表达水平显著下降(P<0.05),且与PDTC呈剂量时间依赖性。结论:PDTC下调NRK细胞中MCP-1mRNA和蛋白的表达。
AIM: To investigate the effects of different concentrations of pyrrolidine dithiocarbamate (PDTC) on the expression of monocyte chemotactic protein-1 (MCP-1) in rat fibroblasts (NRK) under high glucose conditions. Methods: Rat NRK cells were cultured in vitro. Glucose was divided into 6 groups, normal control group, high glucose group 1, high glucose group 2 and glucose group 5.6, 15 and 30mmol / L respectively. The intervention group was glucose 30mmo / L The expression of MCP-1 mRNA in each group was detected by RT-PCR at 24, 48 hours after treatment with PDTC, and the expression of MCP-1 protein in each group was detected by immunocytochemistry (ICC). Results: Compared with the normal control group, the mRNA and protein expressions of MCP-1 in high glucose group were significantly increased (P <0.05), and in a time-dependent manner; while the expression of MCP-1 mRNA and protein was significantly increased after intervention with different concentrations of PDTC Decreased (P <0.05), and with dose-dependent PDTC. Conclusion: PDTC down-regulates the expression of MCP-1 mRNA and protein in NRK cells.
