目的研究白术七物颗粒对结肠慢传输便秘(slow transit constipation,STC)大鼠结肠组织多巴胺D2受体(Drd2)的影响,初步探讨白术七物颗粒对STC的治疗作用及作用机制。方法采用随机数字表法将60只SD大鼠分为空白组、模型组、阳性组、白术七物颗粒低、中、高剂量组,每组各10只,模型组、阳性组、白术七物颗粒低、中、高剂量组予以皮下注射盐酸吗啡注射液建立STC模型,空白组给予等量生理盐水皮下注射。记录造模期间大鼠的一般情况,造模前后各组大鼠24h粪便干重、肠道墨汁推进率。造模成功后,白术七物颗粒低、中、高剂量组大鼠灌服白术七物颗粒水溶液,阳性组大鼠灌服伊托必利片水溶液,模型组、空白组灌服等量等渗生理盐水。给药14d后检测各组大鼠结肠组织Drd2的表达水平。结果空白组大鼠运动灵活,饮食、饮水及大小便正常,其余各组大鼠造模后出现毛发枯黄、懒动、大便次数减少、颗粒粗大质硬等情况。造模前24h 6组大鼠粪便干重比较,差异无统计学意义(P>0.05),造模后,模型组、阳性组、白术七物颗粒低、中、高剂量组大鼠粪便干重、肠道墨汁推进率均低于空白组(P<0.05),模型组、阳性组、白术七物颗粒低、中、高剂量组大鼠结肠组织Drd2表达水平高于空白组(P<0.05),阳性组、白术七物颗粒低、中、高剂量组大鼠结肠组织Drd2表达水平低于模型组(P<0.05),白术七物颗粒低剂量组(白低组)大鼠结肠组织Drd2表达水平与阳性组相比,差异无统计学意义(P>0.05),白术七物颗粒中剂量组(白中组)大鼠、白术七物颗粒高剂量组(白高组)大鼠结肠组织Drd2表达水平均低于阳性组(P<0.05)。结论白术七物颗粒能有效治疗STC,其作用可能与降低结肠组织Drd2的表达水平有关。
Objective To investigate the effect of Baizhuqiwu granule on the expression of Dopamine D2 receptor (DRD2) in colon tissue of slow transit constipation (STC) rats and to explore the therapeutic effect and its mechanism of Baizhuqiwu granule on STC. Methods Sixty Sprague-Dawley rats were randomly divided into blank group, model group, positive group, Atractylodes senticosus particle group in low, medium and high dose groups, 10 in each group, model group, positive group, STC model was established by subcutaneous injection of morphine hydrochloride injection in the low, medium and high dose groups, while the blank group was injected subcutaneously with the same amount of saline. Record the general situation of rats during modeling, before and after modeling in each group of rats 24h dry weight, intestinal ink advancement rate. After the model was established successfully, the rats in low and medium dose group and high dose group were fed with aqueous solution of Atractylodes sesquilecollae. The rats in the positive group were fed with aqueous solution of itopride, and the model group and the blank group were given the same amount of isotonic solution Physiological saline. After administration for 14 days, the expression of Drd2 in the colon tissues of each group was detected. Results The rats in the blank group were flexible in movement, eating, drinking and urine were normal. The rest of the rats in each group showed yellowish hair, lazy movement, reduced stool frequency and coarse and hard granules. There was no significant difference in the fecal weight of rats in the 6th group before modeling (P> 0.05). After modeling, the dry weight of feces in the model group, the positive group and the Atractylodes, (P <0.05). The expression of Drd2 in the model group, the positive group and the Atractylodes Granule group in low, medium and high dose groups was higher than that in the blank group (P <0.05) (P <0.05). The expression of Drd2 in the colon of low, medium and high dose group was lower than that of the model group (P <0.05) (P> 0.05). The rats in Atractylodes Chuanxu Granules medium dose group (Baizhong group), the rats in Atractylodes Chuanxiong granules high dose group (Bai Gao group) had no significant difference in the expression of Drd2 The expression levels were lower than those in the positive group (P <0.05). Conclusion Atractylodes Granule can effectively treat STC, which may be related to the decrease of Drd2 expression in colon.
