目的研究佛波醇酯(PMA)和细胞外信号调节蛋白激酶(ERK1/2)抑制剂PD98059对全反式维甲酸(ATRA)体外抑制人肝癌细胞株(HepG2)表达骨桥蛋白(OPN)的影响及其机制的初步探讨。方法PMA、PD98059分别处理经ATRA处理的肝癌细胞HepG2,光镜下观察细胞形态,West-ernblot检测骨桥蛋白(OPN)、ERK1/2表达及其磷酸化变化,ImagePro软件分析各条带灰度值变化并做统计学分析。结果在ATRA显著抑制肝癌细胞表达OPN并诱导细胞分化的基础上,PMA可刺激OPN表达,这种作用可被PD98059明显抑制。结论激活ERK1/2通路可逆转ATRA抑制HepG2表达OPN的作用。
Objective To investigate the effect of ATRA on the expression of osteopontin (OPN) in HepG2 cells induced by allopurinol (PMA) and PD98059, an inhibitor of extracellular signal-regulated protein kinase (ERK1 / 2) Impact and its mechanism of a preliminary discussion. Methods Hepatoma cell line HepG2 treated with ATRA was treated by PMA and PD98059 respectively. The cell morphology was observed under light microscope. The expression of OPN and ERK1 / 2 was detected by Western-ernblot. The changes of phosphorylation of ERK1 / 2 were analyzed by ImagePro software. Value changes and do statistical analysis. Results PMA stimulated the expression of OPN on the basis of ATRA significantly inhibited the expression of OPN and induced cell differentiation in hepatoma cells. This effect was significantly inhibited by PD98059. Conclusion Activation of ERK1 / 2 pathway can reverse the effect of ATRA on HepG2 expression in OPN.
