以非洲菊3个品种叶片为材料,研究了不同基因型、叶片叶龄和叶片不同部位以及激素对非洲菊愈伤组织诱导和不定芽分化的影响。结果表明:诱导愈伤组织较为适宜的外植体为嫩叶,以MS+6-BA 1.0~5.0 mg/L+NAA 0.2 mg/L为培养基,愈伤组织诱导率均>98%;诱导分化不定芽较为适宜的外植体为叶柄,最适培养基为MS+TDZ 2.0 mg/L+NAA 0.2 mg/L,但不定芽分化率较低,仅为4.2%。
The germination rate and adventitious bud differentiation of Gerbera jamesonii were studied with three genotypes of Gerbera as the materials. The effects of different genotypes, leaf age, different parts of leaves and hormone on the callus induction and adventitious bud differentiation of gerbera jamesonii were studied. The results showed that the more appropriate explants of induced callus were young leaves. The callus induction rate was> 98% with MS + 6-BA 1.0-5.0 mg / L and NAA 0.2 mg / L as medium. The explants suitable for adventitious bud differentiation were petioles. The best medium was MS + TDZ 2.0 mg / L + NAA 0.2 mg / L, but the adventitious bud differentiation rate was only 4.2%.
