目的观察反式-3,5,4'-三甲氧基二苯乙烯(3,5,4'-trimethoxy-trans-stilbene,BTM)对碘乙酸钠致大鼠骨关节炎模型的影响。方法通过对大鼠右侧膝关节内注射碘乙酸钠制备骨关节炎模型,将造模成功的大鼠随机分为模型组、羧甲基纤维素钠溶媒组、反式-3,5,4'-三甲氧基二苯乙烯25、50、100 mg·kg-1剂量组、双醋瑞因胶囊8.0 mg·kg-1阳性对照组;另设正常对照组、假造模组(n=10)。每日1次灌胃给药,连续4周。末次给药后次日,大鼠麻醉腹主动脉取血,分别检测血清中白细胞介素-1β、肿瘤坏死因子-α、基质金属蛋白酶-2水平。采血后处死大鼠,切开右侧膝关节,进行关节腔大体形态学观察及软骨组织病理学检查。结果给药4周后,反式-3,5,4'-三甲氧基二苯乙烯50、100 mg·kg-1剂量组对关节损伤有显著地抑制作用(P<0.05,P<0.01),能改善关节面软骨的损伤;反式-3,5,4'-三甲氧基二苯乙烯25、50、100 mg·kg-1剂量组血清中白细胞介素-1β、肿瘤坏死因子-α含量均显著低于模型组(P<0.05,P<0.01)。结论反式-3,5,4'-三甲氧基二苯乙烯对碘乙酸钠致大鼠骨关节炎有很好的治疗作用,其作用可能与减少炎症因子白细胞介素-1β和肿瘤坏死因子-α有关。
Objective To investigate the effect of trans-3,5,4'-trimethoxy-trans-stilbene (BTM) on the osteoarthritis model induced by sodium iodoacetate in rats. Methods The rat model of osteoarthritis was prepared by injection of sodium iodoacetate on the right knee of rats. The successful rats were randomly divided into model group, sodium carboxymethyl cellulose vehicle group, trans-3,5,4 '- trimethoxy stilbene 25,50,100 mg · kg-1 dose group, diacerein capsules 8.0 mg · kg-1 positive control group; another normal control group, fake model group (n = 10) . Gavage once daily for 4 weeks. The day after the last administration, the rats were anesthetized and the blood was taken from the abdominal aorta to detect the level of interleukin-1β, tumor necrosis factor-α and matrix metalloproteinase-2 respectively. Rats were sacrificed after blood collection, the right knee was dissected, gross morphology of articular cavity and histopathological examination of cartilage were observed. Results After 4 weeks of administration, trans-3,5,4'-trimethoxy-Stilbene 50,100 mg · kg-1 significantly inhibited joint damage (P <0.05, P <0.01) , Can improve the articular cartilage injury; interleukin-1β, tumor necrosis factor-α Content were significantly lower than the model group (P <0.05, P <0.01). CONCLUSION: Trans-3,5,4'-trimethoxy-stilbene has a good therapeutic effect on sodium iodoacetate-induced osteoarthritis in rats, which may be related to the decrease of interleukin-1β and tumor necrosis factor -α related.
