目的研究几种主要热休克蛋白(heat shockprotein,HSP)在不同病理改变的宫颈组织中的表达。方法根据病理诊断,把478例宫颈活检标本分为宫颈癌组(63例)、宫颈上皮内瘤形成组(CIN组,106例)、宫颈炎组(293例)及正常宫颈组(16例)。采用以特异性复合cRNA为内参照的定量RTPCR法,检测各组标本的HSP70、HSP90α和HSP90βmRNA的表达。结果(1)HSP70、HSP90α和HSP90βmRNA在宫颈浸润癌、CIN、宫颈炎及正常宫颈组织中的表达量均呈现阶梯式下降,差异均有极显著意义(P<0.01)。(2)HSP90βmRNA在晚期宫颈癌中的表达比早期表达高(P<0.05);HSP70和HSP90βmRNA在低分化宫颈癌中的表达比高分化表达高(P<0.05);而HSP70、HSP90α和HSP90βmRNA在不同组织类型宫颈癌中的表达均无显著性差异(P>0.05)。结论热休克蛋白在宫颈癌的发生和发展中起着重要的协同作用。HSP70和HSP90α与宫颈癌细胞的转化和增殖密切相关,HSP90β可能参与细胞的分化。
Objective To study the expression of several major heat shock proteins (HSPs) in cervical tissues with different pathological changes. Methods 478 cervical biopsy specimens were divided into cervical cancer group (63 cases), cervical intraepithelial neoplasia group (106 cases), cervicitis group (293 cases) and normal cervical group (16 cases) according to pathological diagnosis. . Quantitative RT-PCR was used to detect the expression of HSP70, HSP90α and HSP90βmRNA in each group. Results (1) The expression levels of HSP70, HSP90α and HSP90βmRNA in cervical cancer, CIN, cervicitis and normal cervical tissue showed a stepwise decline, the differences were significant (P <0.01). (2) The expression of HSP90βmRNA in advanced cervical cancer was higher than that in early stage (P <0.05); The expression of HSP70 and HSP90βmRNA in poorly differentiated cervical cancer was higher than that in poorly differentiated cervical cancer (P <0.05); while HSP70, HSP90α and HSP90βmRNA There was no significant difference in the expression of different types of cervical cancer (P> 0.05). Conclusion Heat shock protein play an important synergistic effect in the occurrence and development of cervical cancer. HSP70 and HSP90α are closely related to the transformation and proliferation of cervical cancer cells, HSP90β may be involved in the differentiation of cells.
