为了揭示黄瓜绿斑驳花叶病毒(Cucumber green mottle mosaic virus,CGMMV)安徽分离物分子变异及系统进化情况,从安徽省5个地区采集感染CGMMV的葫芦科样本。提取感病样本总RNA,经RT-PCR扩增、克隆和测序,获得17个CGMMV分离物的cp基因序列。序列比对发现,17个CGMMV安徽分离物的cp基因核苷酸序列相似性极高,达98.4%~100%,与中国及东亚国家和地区的各个CGMMV分离物cp基因核苷酸序列相似性也非常高,达98.6%~100%,而与CGMMV西班牙分离物和俄罗斯分离物cp基因核苷酸序列相似性相对较低,为90.7%~92.6%。从构建的系统关系树可以看出,17个CGMMV安徽分离物与中国及东亚国家和地区的各个CGMMV分离物形成1个单独的分支,而CGMMV俄罗斯与西班牙分离物聚成另1个分支,说明来源于中国和东亚的CGMMV亲缘关系较近。
In order to reveal the molecular variation and phylogenetic analysis of Cucumber green mottle mosaic virus (CGMMV) in Anhui Province, cucurbits infected with CGMMV were collected from five areas of Anhui Province. The total RNA of susceptible samples was extracted and the cp gene sequences of 17 CGMMV isolates were obtained by RT-PCR amplification, cloning and sequencing. Sequence alignment revealed that the nucleotide sequence of cp gene of 17 isolates of CGMMV was very high (98.4% -100%), which was similar to that of CGMV isolates in China and East Asian countries But also very high, reaching 98.6% -100%. However, the nucleotide sequence of cp gene from CGMMV Spain isolate and Russian isolate was relatively low, ranging from 90.7% to 92.6%. It can be seen from the constructed phylogenetic tree that each of the CGMMV isolates of 17 CGMMVs forms a single branch with each of the CGMMV isolates in China and East Asian countries and that the CGMMV Russian and Spanish isolates form another branch, CGMMV originating from China and East Asia has a relatively close relationship.
