Purification of silymarin from Silybum marianum extracts using macroporous resins

  Enrichment and purification of silymarin from Silybum marianum extracts were performed using five types of macroporous resins.Static tests indicated that HPD-600 resin had the best performance.To optimize the separation process, dynamic adsorption and desorption tests were carried out.The optimal adsorption parameters were as follows: initial silymarin concentration in the sample, 7.64 mg/mL;pH, 5.0;sample loading, 2.3 bed volume (BV);flow rate, 2 BV/h;and temperature, 25℃.The optimal desorption parameters were: deionized water and 20% ethanol, 5 BV each;60% ethanol, 10 BV;and flow rate, 2 BV/h.After HPD-600 purification, silymarin content in the extract increased from 11.38% to 53.41%, and the recovery yield was 82.24%.The results demonstrate that HPD-600 resin can efficiently concentrate silymarin in the S.marianum extracts and may also be used for silymarin isolation from other plants.
