Aims: Previous study from our lab reported that sonic hedgehog (Shh) pathway plays a protective role in myocardial infarction (MI) through against the damage of cardiomyocytes,but the underlying mechanism is still not clear.Methods and results: Our study showed that Shh pathway agonist SAG (2 μ M) increased the expression of LC3-Ⅱ,and induced the formation of autophagosomes in cultured H9c2 cardiomyocytes under oxygen glucose deprivation (OGD) 1 h and 4 h.Shh pathway-induced autophagy was further confirmed by the level of LC3-Ⅱ enhanced by SAG (2 μ M) in the presence of lysosomal protease inhibitors chloroquine (10 μ M) or autophagosome-lysosome fusion inhibitor bafilomycin A1 (10 nM).Inhibition of autophagy by 3-methyladenine (3-MA,5mM) blocked SAG-induced cell survival and apoptosis inhibition under OGD.
