Sugars have long been considered as only a type of energy substance.Recently,people began to realize that sugars are important life substances as Protein and Nucleic Acid.They involved in the recognition process of cells as biological information molecule,and may play a more important role in some physiological activities.In this paper,a novel method was developed for the simultaneous determination Lactate and sugars in the serum of type 2 diabetic rats and health controls.Samples were carded out oximation and silylation reaction before injection [1-3].The chromatography was carried out on a DB-5 fused silica capillary column,and the column temperature was maintained at 70℃for 1 rmin and then programmed from 70 to 300℃ at the rate of 8℃,then hold 3rmin at 300℃.Lactate and 8 sugars were separated perfectly and identified accurately according to their Mass information (The chromatogram was shown in figure.1),where Lactate,Glucose,Fructose,Galactose,1,5-Anhydro-Sorbitol in the serum were quantitatively analyzed by standard curves.There are significant difference between the two groups by using t-test (P<0.05) based on their absolute concentration.Principal components analysis (PCA) method [4,5] was successfully applied to estabfish the discrimination model of both samples also.The results shown that it is helpful for the research ofmetabolomics.
