Background- Local flow patterns determine the uneven distribution of atherosclerotic lesions.Sensing mechanical forces,membrane lipid rafts are crucial for shear stress-regulated endothelial function.To investigate the role of lipid rafts in regulating endothelial function under shear stress,we compared the translocation of proteins in lipid rafts under different flow patterns by quantitative proteomics.Methods and Results- Lipid raft proteins were isolated from endothelial cells (ECs) after 2-hr exposure to pulsatile shear stress (PS,12+/-4 dyn/cm2) or oscillatory shear stress (OS,0.5+/-4 dyn/cm2).
