Gold nanoparticle layer:Regulation of stem cell behaviors and macromolecular delivery

  Gold nanoparticle layer(GNPL)adopts the morphology of three-dimensional micro-and nanosized structures,consisting of gold nanoparticle(GNP)aggregates with different sizes.In our previous studies,we have discovered that GNPL,as a representative material with surface topological structures,has a direct influence on protein adsorption/activity(1,2)and cell behavior including adhesion,spreading,and differentiation(3-5).Here,the influence of surface roughness on the pluripotency of embryonic stem cells(ESCs)and the application of GNPL for macromolecular delivery under laser irradiation were studied.On one hand,GNPLs with nano-and low sub-microscale surface roughness(Rq < 392 nm)are conducive to the long-term maintenance of mESC pluripotency,while GNPLs with high sub-micro-and microscale surface roughness(Rq > 573 nm)result in a significant loss of cell pluripotency and a faster undirectional differentiation(6).
