：
猪繁殖与呼吸综合征病毒(PRRSV)能够导致母猪繁殖性能下降、不孕、流产等,在猪场中往往与其他病原混合感染,给该病毒的诊断带来较大困难.本实验设计合成了一套引物和TaqMan探针,以扩增PRRSV的核衣壳蛋白(N)基因,通过反应条件的优化,建立了快速定量检测PRRSV的荧光定量RT-PCR方法.结果表明该方法具有较高的特异性、重复性和敏感性,对PRRSV细胞培养物的检测下限为0.01 ×TCID5

猪繁殖与呼吸综合征病毒(PRRSV)能够导致母猪繁殖性能下降、不孕、流产等,在猪场中往往与其他病原混合感染,给该病毒的诊断带来较大困难.本实验设计合成了一套引物和TaqMan探针,以扩增PRRSV的核衣壳蛋白(N)基因,通过反应条件的优化,建立了快速定量检测PRRSV的荧光定量RT-PCR方法.结果表明该方法具有较高的特异性、重复性和敏感性,对PRRSV细胞培养物的检测下限为0.01 ×TCID50,敏感性比常规RT-PCR高100倍；对来自皖北地区不同猪场的80份可疑猪血清样本检测,PRRSV阳性率为36％,高于常规RT-PCR检测方法,与ELISA试验的符合率达98％..该方法具有快速、准确、灵敏度高等优点,将为皖北地区各猪场早期检测和防控PRRSV提供重要的基础.
