本试验旨在研究扇贝酱(以扇贝、大米、豆粕为主要原料经发酵而制得)对血管紧张素转化酶活性的抑制作用.以马尿酰-组氨酰-亮氨酸为反应底物,血管紧张素转化酶(ACE)为催化剂,反应所生成的马尿酸为测定指标,未加酶抑制剂的反应为空白对照.采用液相色谱法,选择Amethyat-C18色谱柱(4.6mm×150mm,粒径5μm),柱温25℃,流动相为乙腈-超纯水(体积比为25:75),流速0.8 mL/min

本试验旨在研究扇贝酱(以扇贝、大米、豆粕为主要原料经发酵而制得)对血管紧张素转化酶活性的抑制作用.以马尿酰-组氨酰-亮氨酸为反应底物,血管紧张素转化酶(ACE)为催化剂,反应所生成的马尿酸为测定指标,未加酶抑制剂的反应为空白对照.采用液相色谱法,选择Amethyat-C18色谱柱(4.6mm×150mm,粒径5μm),柱温25℃,流动相为乙腈-超纯水(体积比为25:75),流速0.8 mL/min,检测波长228nm,进样量为10μL.结果显示,在试验测定的样品浓度范围内(0.1-5mg/mL),样品对ACE的抑制作用呈现出由弱到强,而后又由强到弱的变化,最高抑制率为41.30%,对应样品浓度为1mg/mL.研究结果表明,扇贝酱能一定程度的抑制血管紧张素转化酶的活性,可能与其发酵过程中产生了一定量的降血压肽有关;但该活性并不与扇贝酱的浓度呈正相关,可能是其本身含盐含量较高的原因.作用机制有待进一步探讨.
