Histological effect and functional evaluation of adenovirus-mediated BDNF transfer on sciatic nerve

  Objective To observe the peripheral nerve regeneration of injured sciatic nerve after adenovirus-mediated gene transfer of brain-derived neurotrophic factor (BDNF) in adult rats.Methods Based on silicone tube graft as a support to bridge adult rat sciatic nerve gaps, Wistar rat were divided into 3 groups and microinjected with recombinant adenovirus vector of BDNF (AxCA-BDNF), BDNF and simple injection of virus buffer to the sciatic nerve respectively.With transmission electron microscope (TEM), histomorphometry and image analysis, we calculated the number of the new nerve fibers and motoneurons in anterior horn of the spinal cord respectively, and measured the myelin sheath thickness of the new nerve fibers.Using sciatic function index (SFI) and the electrophysiological parameters (including the nerve conduction velocity and compound muscle action potential amplitude), we assessed the adult rat sciatic nerve function recovery following nerve repaire after adenovirus-mediated gene transfer of BDNF.Results Those data were better than those of the control group.The result of retrograde axonal transport of HRP tracer indicated the injured sciatic nerve reconnected with neurons in spinal cord.Conclusion Gene therapy for sciatic nerve injury of adult rats by adenovirus-mediated gene transfer of brain-derived neurotrophic factor in vivo not only promotes nerve regeneration but also protects the neurons in the spinal cord.
