花生EMS高油酸突变体FAD2B基因突变体的鉴定

  运用近红外技术,从油酸含量为44.2%的传统出口型大花生品种鲁丰2号EMS浸种诱变群体中筛选出油酸含量高达64.2%的突变体E2-4-83-12.FAD2基因克隆和序列分析表明,该突变体AD2B基因编码区第313位C碱基突变成T碱基,导致所编码的氨基酸第105为组氨酸突变成酪氨酸.该突变为新突变,其导致的氨基酸变化发生于第一个组氨酸盒内,进一步通过酵母表达系统检测了FAD2B基因活性.突变型FAD2B基因和野生型FAD2B基因酵母表达载体以及空载体pYES2分别导入酿酒酵母INVSC1菌株体内进行诱导表达,通过气相色谱和质谱分析酵母中脂肪酸含量.结果表明,含野生型FAD2B基因的酵母细胞中有亚油酸产生,而含突变型FAD2B基因和空载体的酵母体内无亚油酸产生.无功能的FAD2B基因导致该EMS突变体油酸含量提高.
