目的 探讨新生霉素对伊马替尼敏感及耐药慢性粒细胞白血病细胞的作用及其机制.方法 MTT比色法检测新生霉素对伊马替尼敏感K562及伊马替尼耐药K562/G01细胞的增殖抑制作用;PI染色后流式细胞仪检测新生霉素对伊马替尼敏感和耐药慢性粒细胞白血病细胞细胞周期的影响;利用CFSE染色后流式细胞仪评估新生霉素对K562及K562/G01细胞增殖分裂的抑制作用;利用Annexin V-FITC/PI双染,流式细胞仪检测新生霉素对伊马替尼敏感和耐药慢性粒细胞白血病细胞细胞凋亡的影响;Westemblot检测新生霉素处理48h后细胞周期G2/M期相关调控因子和凋亡相关蛋白的改变.
