目的:总结实体瘤外周血中循环肿瘤细胞(CTC)和转移相关性的研究进展。方法:以“循环肿瘤细胞、实体瘤、转移”为关键词,检索2000-01-2010-10 PubMed、Science Direct、Ovid、Springer、CNKI和维普等数据库的相关文献。纳入标准:关于实体瘤CTC与转移密切相关的分子机制、临床相关性的文献。共纳入分析42篇文献。结果:随着分子生物学和材料技术的发展,越来越多的方法有效地富集和鉴定不同类型实体瘤外周血CTC。细胞基因水平证实CTC具有恶性生物学特性,CTC自身基因和转移相关蛋白谱的表达、肿瘤微环境、免疫系统等因素影响着CTC远处器官转移灶的形成。CTC数目、特定基因、蛋白的表达与治疗疗效、预后等具有相关性。结论:研究CTC参与血液播散转移的机制,为全面、准确地阐明恶性实体瘤转移的机制、个体化的治疗提供新的工具。
Objective: To summarize the research progress of the correlation between circulating tumor cells (CTC) and metastasis in solid tumor peripheral blood. METHODS: The relevant literature of databases such as PubMed, Science Direct, Ovid, Springer, CNKI and VIP was searched using the key words “circulating tumor cells, solid tumors and metastasis”. Inclusion criteria: The molecular mechanisms of solid tumor CTCs closely related to metastasis, clinical relevance literature. A total of 42 papers were analyzed. Results: With the development of molecular biology and material technology, more and more methods are available for enrichment and identification of CTCs in peripheral blood of different types of solid tumors. The level of cellular genes confirmed the malignant biological characteristics of CTC, the expression of CTC’s own genes and metastasis-related protein profiles, tumor microenvironment, immune system and other factors affecting the formation of distant organ metastases of CTC. The number of CTCs, the expression of specific genes and proteins are related to the therapeutic efficacy and prognosis. Conclusion: To study the mechanism of CTC involvement in the dissemination and dissemination of blood provides a new tool for comprehensively and accurately elucidating the mechanism of metastasis of malignant solid tumors and individualized treatment.
