Aim:Although the presence of mesenchymal stem cells in the auricular perichondrium has recently been reported, the molecular mechanism concemed with the control of mesenchymal stem cells has yet to be identified.In this study, we prepared a model stimulated by basic fibroblast growth factor (bFGF) in vivo and attempted to reveal the factors related to the dynamics of mesenchymal stem cells in the auricular perichondrium.Method:Regeneration of the bFGF-stimulated perichondrial cartilage in rabbit ears was chronologically investigated from day one to three months after stimulation.We used immunohistochemistry, western blot and cultured rabbit auricular perichondrial cells.We examined expression of Aquaporin (AQP) 1 using, in addition to immunohistochemistry, a double labeling immunofluorescence method with actin.Result:Proliferation and differentiation to cartilage was observed one week after bFGF stimulation.Co-expression of AQP1 was observed in the cellular processes, where actin filaments were aggregated.Conclusion:AQP1 may become one of the markers of proliferating chondrocyte, suggesting that it is involved in the migration of tissue mesenchymal stem cells/cartilage progenitor cells.
