目的研究丹酚酸B、C组分配伍对肾间质纤维化大鼠肾组织炎症细胞趋化因子CCL2、CCL3的作用及机制。方法50只雄性SD大鼠随机分为正常组、模型组、丹酚酸B组、丹酚酸C组、丹酚酸B和C按1∶1配伍组(简称丹酚酸B+C组)。除正常组外,其余各组均给予单侧输尿管结扎术(UUO)建立肾间质纤维化模型。各治疗组即给予相应药物灌胃治疗。第14天处死大鼠,光镜观察肾组织病理、胶原沉积,全自动生化分析仪检测血清肌酐(Cr)、尿素氮(BUN),Western Blot法检测肾组织单核细胞趋化蛋白-1(CCL2)和巨噬细胞炎症蛋白-3α(CCL3)蛋白表达。结果与模型组比较,各治疗组HE、Masson评分均显著降低,丹酚酸C组血清Cr水平降低明显,各治疗组CCL2、CCL3相对表达显著降低。结论丹酚酸B、C组分配伍可抑制肾组织CCL2和CCL3蛋白表达,从而改善UUO大鼠肾功能及肾脏病理,并对其肾脏起到一定的保护作用。
Objective To study the effect and mechanism of compatibility of Salvianolic acid B and C components on chemokines CCL2 and CCL3 in renal tissue of renal interstitial fibrosis rats. Methods Fifty male Sprague-Dawley rats were randomly divided into normal group, model group, salvianolic acid B group, salvianolic acid C group, salvianolic acid B and C according to 1: 1 compatibility group (salvianolic acid B + C group) . In addition to the normal group, the other groups were given unilateral ureteral ligation (UUO) to establish a model of renal interstitial fibrosis. Each treatment group is given the corresponding drug gavage treatment. The rats were sacrificed on the 14th day, and the pathological changes of renal tissue were observed under light microscope. The serum creatinine (Cr) and urea nitrogen (BUN) were detected by automatic biochemical analyzer. The levels of MCP- CCL2) and macrophage inflammatory protein-3 alpha (CCL3) protein expression. Results Compared with the model group, the HE and Masson scores of all the treatment groups were significantly decreased, while the levels of serum Cr in salvianolic acid C group were significantly decreased. The relative expression of CCL2 and CCL3 in each treatment group was significantly decreased. Conclusion Combination of salvianolic acid B and C can inhibit the expression of CCL2 and CCL3 protein in renal tissue and thus improve renal function and renal pathology in UUO rats and play a protective role on their kidneys.
