In order to find new safer antibacterial compound, it was determined by disc diffusion method on the antibacterial activity and action mechanisms of clausenamide and curcumin to 4 pathogenic bacteria in vitro. The results indicated that clausenamide has broader spectrum and higher antibacterial activity than curcumin against the tested bacteria Escherichia coli AS1.1068, Staphylococcus aureus ATCC25923, Bacillus subtilis and Salmonella lignieres1900.The diameters of inhibition zone of clausenamide against 4 standard strains(S.aureus, B.subtilis, S.lignieres and E.coli) at the concentration of 32 mg/ml, were 12.67 mm, 10.33 mm ,11.17 mm and 6.67mm, but the curcumin were 6.00mm, 9.12mm, 7.50mm and 6.00mm, respectively. The minimum inhibitory concentration (MIC)were measured by broth dilution method at the concentrations ranging from 2.00 to 0.125 mg/mL.The MIC values of clausenamide were 0.5mg/ml against S.aureus.SDS-polyacrylamide gel electrophoresis showed that in its role, the expression of S.aureus decreased in total protein content, indicating that the inhibitory mechanism may be destructive role of bacteria in the protein or inhibit its synthesis.
