The nuclease hypersensitivity element (NHE) Ⅲ1 upstream of the P1 promoter that controls 85-90% of the human c-myc promoter transcriptional activation has been attracted considerable research over the past two decades1-2.The NHE Ⅲ1 which is a 27-base-pair sequence, located-142 to -115 base pairs upstream of the P1 promoter, comprises 5 sets of G-tracts, while only four G-tracts of shorter c-myc oligonucleotide can fold into a distinct G-quadruplex by NMR spectroscopy3.The G-quadruplex formed from the four G-tracts at the 3 end is considered to be biologically relevant on the basis of that a ligand can selective interaction with it will induce a decrease in c-myc transcription4.
