The classical method of human pluripotent stem cell (hPSC) differentiation occurred via embryoid body (EB) formation,but the most population methodology should be that some functional somatic cells can be generated without differentiation via EB formation from hPSC.The difference between hPSC differentiation with and without EB was not clear.In this study,we used RT-PCR to analyze changes in the expression of genes that are involved in the directional preference of hPSC differentiation and epithelial to mesenchymal transition (EMT) associated markers.Additionally,microarrays were used to survey the differences and clustering for the directional differentiation of hPSCs and the committed cell biology mechanism.We found that both EMT and the cell interaction and communication patterns were altered by the two methods.Furthermore,these changes were due to the different directions of hPSC differentiation.These findings may be helpful for the lineage-specific committed differentiation of hPSCs.
