In higher plants,overexpression of miR156 prolongs the juvenile phase of development,delays flowering,promotes tillering,increases plant biomass yield via target the SQUAMOSA PROMOTER BINDING PROTEIN-LIKE(SPL)family genes.Recent ten years,strigolactones(SLs),a novel class of terpenoid plant hormones,were found act as endogenous signals to inhibit shoot branching in plants.Although the microRNAs and miR156-target SPL gene family had been identified in Salvia miltiorrhiza,a model medicinal plant,there are no reports to date about functions ofmicroRNAs in S.miltiorrhiza.Here,we isolateda miR156 precursor sequence(Sm-MIR156a)in S.miltiorrhiza; RT-qPCR showed that Sm-MIR156a expresses higher level in stem and its expression level gradually declines along with the growth of S.miltiorrhiza; Overexpression of Sm-MIR156a in Nicotiana benthamianaresults in obvious morphological alterations,including young state and more branches; The content of nicotine,caffeic acid and scopoletin in transgenic line OE-1reduced to 47%,30%and 29%of control,respectively;In OE1,the expression level of CCD7,CCD8 andBCR1,the enzyme genes of SLs synthesis pathway,decreased to 25%,49%and 67%of control,respectively.
