Purpose: Reversible immortalization holds great potential for primary tissue expansion to develop cell-based therapies as well as for basic research.Primary mouse dental papilla cells (mDPCs), as precursors of odontoblasts, are promising candidates for odontoblastic differentiation.Due to their limited lifespan and low proliferation activity, however, primary mDPCs are of limited use for basic research.Here we explore a way to generate a tamoxifen-mediated reversible immortalization mDPCs cell line.Methods:mDPCs were isolated from dental papilla of E18.5 mouse mandibular first molar.Lentivirus veetor carrying SV40T antigen (Tag) flanked by loxP sites was used to immortalize mDPCs.
