Objective Huntingtin-associated protein-1 (HAP1) is the first protein identified to interact with the N-terminus of huntingtin(htt).However, the function of HAP 1 remains unclear.The present study is to explore the effect of HAP1 on the membrane transport of KV2.1 in PC12 cells.Methods SiRNAs (small interfering RNAs) technique was used to silence the expression of HAP1 in PC12 cells.Potassium current was recorded by whole-cell patch clamp technique and RT-PCR was used to observe the effect of HAP 1 silencing on the mRNA expression of Kv2.1 in PC 12 cells.Immunofluorescence technique was used to observe the effects of HAP1 protein on Kv2.1 potassium channel cytoplasmic expression and membrane transport in PC 12 cells.Results (1) The silence of HAP1 inhibited the voltage-sensitive potassium current and induced slightly up-regulation of the Kv2.1 mRNA expression in PC 12 cells.(2) The silence of HAP1 increased the cytoplasmic expression of Kv2.1 but inhibited the membrane transport of KV2.1.Conclusion HAP1 can regulate the membrane transport of KV2.1 expressed in PC12 cells.
