尼罗罗非鱼(Oreochromis niloticus)dmrt6基因分离及其功能

  目前dmrt6(double sex-and-Mab-3-related transcription factor 6)基因只报道存在于四足类(Tetrapod)和矛尾鱼(Latimeria chalumnae)基因组中,且对其进化和功能鲜有研究.本研究从鲈形目(Perciformes,五种丽鱼和三棘刺鱼)、鲶形目(Siluriformes,南方鲶)和雀鳝目(Siluriformes,斑点雀鳝)分离得到该基因,共线性和系统发生分析均表明在真骨鱼与其它硬骨鱼分歧后dmrt6发生了基因转座.组织分布显示dmrt6在尼罗罗非鱼(Oreochromis niloticus)精巢中高表达,在脑、肝脏、肾和头肾中也存在少量表达,性腺个体发生显示,dmrt6从90 dah(day after hatching)到成体高表达于精巢,而在卵巢中不表达.原位杂交显示dmrt6表达于精巢精母细胞.采用CRISPR/Cas9系统敲除XY罗非鱼Dmrt6后,120 dah组织学、免疫组化分析和激素水平检测显示,对照组已出现各级生精细胞,敲除组性腺精母细胞缺失、Cyp11b2表达下调及11-ketotestosterone水平降低.在150 dah实验组性腺观察到精母细胞和180 dah时出现精细胞,且cyp11b2表达和11-ketotestosterone水平逐渐恢复到对照水平.Dmrt6敲除后可育并能稳定遗传到后代.本研究表明,dmrt6也存在于真骨鱼在内的硬骨鱼中,在多种硬骨鱼中没找到可能是由于测序不完整或次生性丢失,另外,该基因高表达于罗非鱼精母细胞并且敲除后会导致精子发生推迟.
