目的:观察内皮素(ET-1)和肿瘤坏死因子(TNF-α)在急性低压缺氧大鼠肾组织中的表达,探讨低压缺氧条件下肾损伤的机制.
方法:雄性Wistar大鼠16只,随机分为地面组和8.0km组.将动物置于小动物低压舱内,模拟上升至8.0km高度,暴露30min,然后取标本.用ELISA方法测量血清中活性氧(nOS)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和丙二醛(MDA)的水平,并用免疫组织化学方法检测肾脏中ET-1和TNF-α的表达.
结果:与地面组比较,8.0km组大鼠血清中ROS水平显著降低(P<0.01),MDA含量明显增高(P<0.01),SOD和CAT活性不变.免疫组织化学染色显示,ET-1和TNF-α表达均明显增强(P<0.05).
结论:ET-1及TNF-α参与了缺氧性肾损伤的病理过程.
