全氟化合物(Perfluorinated chemicals,PFCs)是一类新型有机污染物,其环境污染问题已经引起了人们的重视.全氟辛酸(perfluorootanoic acid,PFOA)是环境中出现的最典型的PFCs之一,同时又是多种PFCs在环境中的最终转化产物,其在环境中的行为越来越受到广泛关注.相关研究表明,PFOA可以引起生物多种毒性,但其对环境微生物的影响的研究报道较少.本实验以E.coli为模型细菌,对其在PFOA胁迫作用下蛋白质表达水平的变化进行了研究,SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)实验结果表明, E.coli在PFOA的胁迫下,菌体能生成新的蛋白.应用差异蛋白质组学的方法,采用双向电泳技术(2-DE)和质谱分析技术(MS)共检测到43个差异蛋白点,表明E.coli在PFOA的胁迫下,与胁迫响应相关的蛋白种类发生了变化.这些蛋白质参与的生物学过程有待于进一步研究.
