Nowadays,drugs of abuse has spread all over the world and become a serious social problem in every country.Drug testing provides the judicial evidence of illicit drug use by detection the presence of the parent drug and its metabolite in biological matrices.In general,urine testing is most common method to monitor drugs of abuse as urine samples are simple,non-invasive to collect,and available in relatively large quantities.It often used immunoassay for screening assay and gas chromatography mass spectrometry(GC–MS)for the confirmation assay in routine laboratory.However,due to the popularity of club drugs among the young race recently,the more advantageous assay is greatly desired.Hair had received obviously considerable alternative specimen because of its longer detection window(months to years).It can provide a retrospective calendar of an individuals drug use by segmental analysis.However,drug concentration in hair specimens was lower than in the urine,thus the method developments of reduced-size sample analysis and trace concentration drug detection were limited.In our previous study,the simultaneous quantification method of abused drugs in hair has been employed by GC–MS in the selected ion monitoring(SIM)mode1.Although hair testing using GC–MS is an adequate method that provides low cost,unambiguous identification with sufficient sensitivity,the development of simultaneous analytic method was limited that none ideal derivatization can suitable for all drugs and their metabolites.Also,time-consuming derivatization is usually required that limit the throughput.The purpose of this study is to develop next generation of hair testing for drugs of abuse.A rapid method was developed for drug testing in hair by using microwave-assisted extraction(MAE)2,derivatization enhanced approach2 and liquid chromatography tandem mass spectrometry(LC–MS/MS)for simultaneously quantify amphetamines,ketamine,opiates and metabolites.The merits and limitations of derivatization approach are discussed.This method was proved for the practical drug testing in hair,especially for multiple drug abuser samples detection in a single run.
