目的观察中链甘油三酯(medium-chain triglyceride,MCT)对高胆固醇血症小鼠体质量、体脂、血清脂质水平及其体内胆固醇逆转运的影响。方法建立高胆固醇血症小鼠模型,将30只造模成功小鼠随机分为中链甘油三酯组(MCT组)和长链甘油三酯(long-chain triglyceride,LCT)组,给予含2%MCT和2%LCT高脂高胆固醇饲料喂养3个月后,腹腔注射富含3H胆固醇的RAW264.7巨噬细胞悬液,单笼饲养3 d,收集每天粪便,腹主动脉取血,测量体长、肝及附睾脂肪组织。测定血清脂质,用液闪计数仪测定血清、肝及粪便中3H胆固醇放射活性。结果实验结束时,与LCT组相比,MCT组小鼠空腹体质量、体质量增加量、Lee氏指数、附睾脂肪重量及血清TC水平显著降低(P<0.05);两组血清HDL-C、n-HDL-C和TG差异无统计学意义(P>0.05);与LCT组比较,MCT组血清和肝3H胆固醇水平均显著降低(P<0.05),同时48 h粪便中3H胆固醇流出量及72 h总量显著增加(P<0.05)。结论 MCT能够减轻高胆固醇血症小鼠体质量,减少体脂肪积累和改善胆固醇代谢,且能促进体内胆固醇的逆转运。
Objective To investigate the effects of medium-chain triglyceride (MCT) on body mass, body fat, serum lipid and cholesterol transport in hypercholesterolemic mice. Methods The model of hypercholesterolemia in mice was established. Thirty successful model mice were randomly divided into medium-chain triglyceride group (MCT group) and long-chain triglyceride (LCT group) After 3 months of feeding high-fat and high-cholesterol diet with 2% LCT and 2% LCT, RAW264.7 macrophages enriched in 3H cholesterol were injected intraperitoneally, and kept for 3 days in a single cage. Blood samples were taken from the feces and abdominal aorta every day. Body length, liver and epididymal adipose tissue. Serum lipids were measured and radioactivity of 3H cholesterol in serum, liver and feces was measured using a liquid scintillation counter. Results At the end of the experiment, compared with the LCT group, the fasting body weight, body weight gain, Lee’s index, epididymal fat mass and serum TC level in MCT group were significantly decreased (P <0.05). The serum HDL-C, There were no significant differences in n-HDL-C and TG (P> 0.05). Compared with LCT group, serum and liver 3H cholesterol levels in MCT group were significantly decreased (P <0.05) The total amount of 72 h increased significantly (P <0.05). Conclusion MCT can reduce the body mass of hypercholesterolemic mice, reduce body fat accumulation and improve cholesterol metabolism, and can promote the reverse cholesterol transport in vivo.
