Interaction between the actin cytoskeleton and the cadherin/catenin complex in regulating dendritic

  As dendrites are the primary sites through which neurons receive inputs from contacting axons,dendritic development and arborization are critical to neural circuit formation and synaptic connectivity.We have previously shown that the cadherin/catenin complex is critical for activity-dependent dendritic morphogenesis.Here we further demonstrate that inter-neuronal cadherin-cadherin interactions play an important role in regulating dendritic arborization,as expression of the extracellular domain of N-cadherin in soluble form reduces dendritic arborization and blocks the effects of neural activity,as well as over-expression of beta-catenin or alphaN-catenin on increasing dendritic branching.In addition,we show the ability of alphaN-catenin to bind to the actin cytoskeleton is required for regulation of dendritic arbor development by the cadherin/catenin complex,an effect that cannot be mimicked by the equivalent domain of vinculin.Finally,we show using latnmculin A treatment that actin dynamics is required for enhancement but not maintenance of dendritic arbors.
