Transcription factor NF-B plays a pivotal role in many aspects of human cancer.Two major NF-B regulation pathways have been defined: the canonical pathway and the alternative pathway.A primary step in the alternative pathway of NF-B activation requires NF-B2/p 100 processing to generate p52.In most cases, stimuli-induced p 100 processing is dependent on NF-B-indueing kinase (NIK)/IB kinase (IKK)-mediated phosphorylation and ubiquitination.Here we report that posttranslational modification ofpl00 at specific sites by the small ubiquitin-like modifier (SUMO) is another determining factor for stimuli-induced pl00 processing.Our results indicate that a threshold of basal p 100 sumoylation creates a privileged pool of p 100 that are competent for stimuli-induced phosphorylation and subsequent-TrCP recruitment, polyubiquitination and ultimate p52 generation and RelB nuclear translocation.
