目的:建立丹皮炭的HPLC指纹图谱分析方法,为整体控制和评价丹皮炭的质量提供依据。方法:采用HederaODS-3柱(4.6 mm×150 mm,5μm),流动相甲醇-乙腈(1∶1)-0.5‰三氟乙酸水,梯度洗脱,检测波长258 nm。以丹皮酚为参照物,对10批丹皮炭进行指纹图谱的分析。结果:建立了丹皮炭饮片的HPLC指纹图谱共有模式,标定了11个共有峰,10批炭品的相似度在0.830~0.991。结论:该指纹图谱方法简便、准确、重复性好,可专属性地研究丹皮炭的指纹图谱,为有效地控制该饮片的内在质量提供了科学依据。
Objective: To establish a method for HPLC fingerprinting analysis of Danpi charcoal, and provide basis for the overall control and evaluation of the quality of Danpi charcoal. METHODS: A Hedera ODS-3 column (4.6 mm×150 mm, 5 μm) was used. The mobile phase consisted of methanol-acetonitrile (1∶1)-0.5‰ trifluoroacetic acid and the gradient elution was carried out at a detection wavelength of 258 nm. Using paeonol as a reference, fingerprints of 10 batches of charcoal were analyzed. RESULTS: A common model of HPLC fingerprints was established for the Dan Pi Carbonized Pieces and 11 common peaks were calibrated. The similarity of 10 batches of charcoal products was between 0.830 and 0.991. Conclusion: The fingerprinting method is simple, accurate, and reproducible. The fingerprint of Danpi charcoal can be specifically studied, which provides a scientific basis for effectively controlling the intrinsic quality of the decoction.
